The wide-spread interest in the very remarkable specimen of the giant squid, now lying on the beach a few miles below the city, is mainly due to its enormous size. Despite this success, almost half the Boer fighting strength, around 15,000 men, were still in the field fighting by May 1902. However, Kitchener's tactics were costly: Britain was running out of time, patience, and money needed for the war. The British offered terms of peace on various occasions, notably in March 1901, but all them were rejected by Botha and the "Bitter-enders" among the Boers, who pledged to fight until the bitter end and rejected the demand for surrender or any compromise made by the "Hands-uppers". Their reasons included their hatred of the British, loyalty to their dead comrades, solidarity with fellow Boer commandos, a desire for independence, religious arguments, and fear of captivity or punishment. == Disorder and disease == Intrinsically unstructured proteins have been implicated in a number of diseases. Aggregation of misfolded proteins is the cause of many synucleinopathies and toxicity as those proteins start binding to each other randomly and can lead to cancer or cardiovascular diseases. Thereby, misfolding can happen spontaneously because millions of copies of proteins are made during the lifetime of an organism. The aggregation of the intrinsically unstructured protein α-synuclein is thought to be responsible. The structural flexibility of this protein together with its susceptibility to modification in the cell leads to misfolding and aggregation. Genetics, oxidative and nitrative stress as well as mitochondrial impairment impact the structural flexibility of the unstructured α-synuclein protein and associated disease mechanisms. Many key tumour suppressors have large intrinsically unstructured regions, for example p53 and BRCA1. These regions of the proteins are responsible for mediating many of their interactions. Taking the cell's native defense mechanisms as a model drugs can be developed, trying to block the place of noxious substrates and inhibiting them, and thus counteracting the disease.

Some longitudinal incisions of 2mm of depth have to be made through the skin to proceed to the extraction of latex, which can be collected in solid form some minutes after the extraction. The proteases present in the latex of the fruit are inactive precursors that are activated once the papaya is wounded. In 0.3 ml of latex there are about 15 mg of chymopapain. If we want to conserve the proteolytic properties, latex has to be preserved with sodium metabisulfite and stored at a low temperature of about -10 °C. The supernatant is discarded and the ion exchange chromatography can be carried out, with a linear gradient of 100mM (Na+) and different volumes of elution.
