Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Reversed-phase high-performance liquid chromatography is the routine method for purity assessment. Peptides absorb near 214 nm because of the peptide bond, and a gradient of acetonitrile in water separates the intact peptide from deletion sequences, oxidised products, and earlier-eluting fragments at neutral pH. Electrospray ionisation mass spectrometry provides an orthogonal check: the measured mass must agree with the theoretical value. Amino acid analysis and peptide mapping confirm structure but are used less often. Reference standards remain scarce because the peptide is not described in any pharmacopoeia.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.

No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
Lyophilised melanotan-2 is comparatively robust when kept dry, cold and dark, and a desiccated powder stored at minus twenty degrees Celsius or below is generally expected to retain its chemical integrity for extended periods. In solution the peptide is far less stable, with degradation proceeding through oxidation of tryptophan and histidine residues, hydrolysis adjacent to the lactam bridge, and aggregation at higher concentrations. Repeated freeze-thaw cycling accelerates loss of the parent peak. Working aliquots are therefore prepared once, held cold, and used without letting the stock return to ambient temperature.
Quality assessment of research-grade peptide rests mainly on reversed-phase high-performance liquid chromatography for purity and on mass spectrometry for identity confirmation. A single main peak above a stated threshold, commonly ninety-eight percent by peak area, is the usual release criterion applied by suppliers. Independent analyses commissioned by laboratories and consumer organisations have repeatedly reported discrepancies between label claims and measured content, including truncated sequences, residual trifluoroacetate, and lower-than-declared peptide mass. Those findings do not establish that every supplier is unreliable, but they indicate that purity figures printed on a vial are claims requiring verification rather than settled facts.
In the early 2000s, White Castle tried expanding into the Kansas City market, with at least one location in Kansas, but those restaurants were closed several years later. In April 2020, White Castle responded to the COVID-19 pandemic by announcing that the chain would be delivering free meals to healthcare workers. White Castle also offered a free dessert in the month of May 2021 to anyone who showed a vaccination certificate. In certain regions, White Castle offered four hours of paid time off for workers who get both doses of an eligible COVID-19 vaccination and present their vaccination certificate to management.
The first FDA-approved therapeutic monoclonal antibody was a murine IgG2a CD3 specific transplant rejection drug, OKT3 (also called muromonab), in 1986. This drug found use in solid organ transplant recipients who became steroid resistant. Hundreds of therapies are undergoing clinical trials. As of 2025, there were over 200 antibodies approved by the FDA, with about 40% of them targeting oncological targets and 35% for autoimmune and inflammatory diseases. The majority of these are conventional antibodies which make up over 70% of FDA approved antibodies. Most are part of the IgG1 subclass, again making up over 70% of antibodies.
=== Occurrence in bacteria === Hydrogen cyanide is produced by various soil bacteria, including cyanobacteria and representatives of the genera Aeromonas, Bacillus, and Pseudomonas. Biosynthesis proceeds from glycine. A group of alkanenitriles was isolated from Pseudomonas veronii: dodecannitrile, tridecannitrile, tetradecanenitrile, pentadecannitrile, and hexadecannitrile, as well as compounds of similar chain length containing a double bond. From Micromonospora echinospora, structurally related compounds were also isolated, differing by a terminal methyl branch, a double bond, or both. A cyanohydrin containing a phosphonic acid moiety is known from Streptomyces regensis. The aetokthonotoxin from the cyanobacteria Aetokthonos hydrillicola is a brominated indole derivative bearing a nitrile group. It is a potent neurotoxin that frequently causes mortality in bald eagles that ingest it.
== Impact on evolutionary theory == Neutral mutation has become a part of the neutral theory of molecular evolution, proposed in the 1960s. This theory suggests that neutral mutations are responsible for a large portion of DNA sequence changes in a species. For example, bovine and human insulin, while differing in amino acid sequence are still able to perform the same function. The amino acid substitutions between species were seen therefore to be neutral or not impactful to the function of the protein. Neutral mutation and the neutral theory of molecular evolution are not separate from natural selection but add to Darwin's original thoughts. Mutations can give an advantage, create a disadvantage, or make no measurable difference to an organism's survival. A number of observations associated with neutral mutation were predicted in neutral theory including: amino acids with similar biochemical properties should be substituted more often than biochemically different amino acids; synonymous base substitutions should be observed more often than nonsynonymous substitutions; introns should evolve at the same rate as synonymous mutations in coding exons; and pseudogenes should also evolve at a similar rate. These predictions have been confirmed with the introduction of additional genetic data since the theory’s introduction.

In Iran, dill is known as 'shevid' and sometimes, is used with rice and called 'shevid-polo'. It also is used in Iranian 'aash' recipes, and similarly, is called sheved in Persian. In India, dill is known as 'Sholpa' in Bengali, shepu (शेपू) in Marathi, sheppi (शेप्पी) in Konkani, savaa in Hindi, or soa in Punjabi. In Telugu, it is called 'Soa-kura' (herb greens). It also is called sabbasige soppu (ಸಬ್ಬಸಿಗೆ ಸೊಪ್ಪು) in Kannada. In Tamil it is known as sada kuppi (சதகுப்பி). In Malayalam, it is ചതകുപ്പ (chathakuppa) or ശതകുപ്പ (sathakuppa). In Sanskrit, this herb is called shatapushpa. In Gujarati, it is known as suva (સૂવા). In India, dill is prepared in the manner of yellow 'moong dal', as a main-course dish. It is considered to have very good antiflatulent properties, so it is used as 'mukhwas', or an after-meal digestive. Traditionally, it is given to mothers immediately after childbirth. In the state of Uttar Pradesh in India, a small amount of fresh dill is cooked along with cut potatoes and fresh fenugreek leaves (Hindi आलू-मेथी-सोया). In Manipur, dill, locally known as pakhon, is an essential ingredient of chagem pomba – a traditional Manipuri dish made with fermented soybean and rice. In Laos and parts of northern Thailand, dill is known in English as Lao coriander (Lao: ຜັກຊີ or Thai: ผักชีลาว), and served as a side with salad yum or papaya salad. In the Lao language, it is called 'phak see', and in Thai, it is known as 'phak chee Lao'.

Geddy Lee's high-register vocal style has always been a signature of the band–and sometimes a focal point for criticism, especially during the early years of Rush's career when his vocals were high-pitched, with a strong likeness to other singers like Robert Plant of Led Zeppelin. A review in The New York Times opined that Lee's voice "suggests a munchkin giving a sermon". Although his voice has softened, it is often described as a "wail". His instrumental abilities, on the other hand are rarely criticized. He has cited Jeff Berlin, Jack Casady, John Entwistle, Jack Bruce and Chris Squire as being the bassists who had the biggest impact on his playing style. Lee's style, technique, and ability on the bass guitar have been influential to rock and heavy metal musicians, inspiring players including Steve Harris, John Myung, Les Claypool, and Cliff Burton. Lee is able to operate various pieces of instrumentation simultaneously during concerts, most evidently when he plays bass and keyboards, sings, and triggers foot pedals as in the song "Tom Sawyer".
On 3 January 1945, the 82nd Airborne Division conducted a counterattack. On the first day's fighting the 82nd Airborne overran the 62nd Volksgrenadiers and the 9th SS Panzer's positions capturing 2,400 prisoners. The 82nd Airborne suffered high casualties in the process. The attached 551st Parachute Infantry Battalion was all but destroyed during these attacks. Of the 826 men who went into the Ardennes, only 110 came out. Having lost its charismatic leader Lt. Colonel Joerg, and almost all its men either wounded, killed, or frostbitten, the 551 was never reconstituted. The few soldiers who remained were later absorbed into units of the 82nd Airborne. After several days of fighting, the destruction of the 62nd Volksgrenadiers, and what had been left of the 9th SS Panzer Division was complete. For the 82nd Airborne Division the first part of the Battle of the Bulge had ended.

Historians have noted that Chinese scholars wrote these studies instead of "books of prescriptions or advice manuals." In their historical and environmental understanding, no two illnesses were alike, so the healing strategies of the practitioner were unique every time to the specific diagnosis of the patient. Medical case studies existed throughout Chinese history, but "individually authored and published case history" was a prominent creation of the Ming dynasty. An example of such case studies would be the literati physician, Cheng Congzhou, collection of 93 cases published in 1644.
Theodosius II (r. 408–450) formalised Roman law by appointing five jurists as principal authorities and compiling legislation issued since Constantine's reign into the Codex Theodosianus. This process culminated in the Corpus Juris Civilis under Justinian I (r. 527–565), who commissioned a complete standardisation of imperial decrees since Hadrian's time and resolved conflicting legal opinions of the jurists. The result became the definitive legal authority. This body of law covered civil matters and also public law, including imperial power and administrative organisation. After 534, Justinian issued the Novellae (New Laws) in Greek, which marked a transition from Roman to Byzantine law. Legal historian Bernard Stolte distinguishes Roman law as this because Western Europe inherited law through the Latin texts of the Corpus Juris Civilis only. Zachary Chitwood argues that the Corpus Juris Civilis was largely inaccessible in Latin, particularly in the provinces. Following the 7th-century Arab conquests, people began questioning the development and application of law, leading to stronger ties between law and Christianity. This context influenced Leo III (r. 717–741) to develop the Ekloge ton nomon, which placed an emphasis on humanity. The Ekloge inspired practical legal texts like the Farmers' Law, Seamen's Law, and Soldiers' Law, which Chitwood suggests were used daily in the provinces as companions to the Corpus Juris Civilis.
== Discovery == Adiponectin was first characterised in 1995 in differentiating 3T3-L1 adipocytes (Scherer PE et al.). In 1996 it was characterised in mice as the mRNA transcript most highly expressed in adipocytes. In 2007, adiponectin was identified as a transcript highly expressed in preadipocytes (precursors of fat cells) differentiating into adipocytes. The human homologue was identified as the most abundant transcript in adipose tissue. Contrary to expectations, despite being produced in adipose tissue, adiponectin was found to be decreased in obesity. This downregulation has not been fully explained. The gene was localised to chromosome 3q27, a region highlighted as affecting genetic susceptibility to type 2 diabetes and obesity. Supplementation by differing forms of adiponectin was able to improve insulin control, blood glucose and triglyceride levels in mouse models. The gene was investigated for variants that predispose to type 2 diabetes. Several single nucleotide polymorphisms in the coding region and surrounding sequence were identified from several different populations, with varying prevalences, degrees of association and strength of effect on type 2 diabetes. Berberine, an isoquinoline alkaloid, has been shown to increase adiponectin expression, which partly explains its beneficial effects on metabolic disturbances. Mice fed the omega-3 fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) have shown increased plasma adiponectin. Curcumin, capsaicin, gingerol, and catechins have also been found to increase adiponectin expression.

=== Aortic stenosis === Treatment of aortic stenosis is not necessary in asymptomatic patients, unless the stenosis is classified as severe based on valve hemodynamics. Both asymptomatic severe and symptomatic aortic stenosis are treated with aortic valve replacement (AVR) surgery. AVR surgery can be performed using mechanical or tissue valves depending on age and other relevant factors. Trans-catheter Aortic Valve Implantation (TAVI) is an alternative to AVR and is recommended in high risk patients who may not be suitable for surgical AVR. Any angina is treated with short-acting nitrovasodilators, beta-blockers and/or calcium blockers, although nitrates can drastically decrease blood pressure in patients with severe aortic stenosis and are therefore contraindicated. Any hypertension is treated aggressively, but caution must be taken in administering beta-blockers. Any heart failure is treated with digoxin, diuretics, nitrovasodilators and, if not contraindicated, cautious inpatient administration of ACE inhibitors. Moderate stenosis is monitored with echocardiography every 1–2 years, possibly with supplementary cardiac stress test. Severe stenosis should be monitored with echocardiography every 3–6 months.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Low temperature slows hydrolysis and oxidation, the two main routes by which the peptide backbone and side chains are modified. A lyophilised powder stored at −20 °C is more stable than one kept at room temperature, and once the material is dissolved the degradation rate rises, making refrigeration more important.
