Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.

Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Deficiency of leptin has been shown to alter brain proteins and neuronal functions of obese mice that can be restored by leptin injection Leptin receptor signaling in the hippocampus enhances learning and memory Treatment with leptin has been shown to enhance learning and memory in animal models. In humans, low circulating plasma leptin has been associated with cognitive changes associated with anorexia, depression, and Alzheimer's Disease Studies in transgenic mouse models of Alzheimer's disease have shown that chronic administration of leptin can ameliorate brain pathology and improve cognitive performance, by reducing b-amyloid and hyperphosphorylated Tau, two hallmarks of Alzheimer's pathology. Generally, leptin is thought to enter the brain at the choroid plexus, where the intense expression of a form of leptin receptor molecule could act as a transport mechanism. Increased levels of melatonin causes a downregulation of leptin, however, melatonin also appears to increase leptin levels in the presence of insulin, therefore causing a decrease in appetite during sleeping. Partial sleep deprivation has also been associated with decreased leptin levels. Mice with type 1 diabetes treated with leptin or leptin plus insulin, compared to insulin alone had better metabolic profiles: blood sugar did not fluctuate so much; cholesterol levels decreased; less body fat formed.
=== Content protection === DisplayPort 1.0 includes optional DPCP (DisplayPort Content Protection) from Philips, which uses 128-bit AES encryption. It also features full authentication and session key establishment. Each encryption session is independent, and it has an independent revocation system. This portion of the standard is licensed separately. It also adds the ability to verify the proximity of the receiver and transmitter, a technique intended to ensure users are not bypassing the content protection system to send data out to distant, unauthorized users. DisplayPort 1.1 added optional implementation of industry-standard 56-bit HDCP (High-bandwidth Digital Content Protection) revision 1.3, which requires separate licensing from the Digital Content Protection LLC. DisplayPort 1.3 added support for HDCP 2.2, which is also used by HDMI 2.0.
Aminoglycosides display concentration-dependent bactericidal activity against "most gram-negative aerobic and facultative anaerobic bacilli" but not against gram-negative anaerobes and most gram-positive bacteria. They require only short contact time, and are most effective against susceptible bacterial populations that are rapidly multiplying. These activities are attributed to a primary mode of action as protein synthesis inhibitors, though additional mechanisms are implicated for some specific agents, and/or thorough mechanistic descriptions are as yet unavailable. These agents exhibit a post-antibiotic effect in which there is no or very little drug level detectable in blood, but there still seems to be inhibition of bacterial re-growth. This is due to strong, irreversible binding to the ribosome, and remains intracellular long after plasma levels drop, and allows a prolonged dosage interval. Depending on their concentration, they act as bacteriostatic or bactericidal agents.

Howard Walter Florey, Baron Florey of Adelaide and Marston (; 24 September 1898 – 21 February 1968) was an Australian pharmacologist and pathologist who shared the Nobel Prize in Physiology or Medicine in 1945 with Ernst Chain and Sir Alexander Fleming "for the discovery of penicillin and its curative effect in various infectious diseases". Although Fleming received most of the credit for the discovery of penicillin, it was Florey and his team at the University of Oxford who made it into a useful and effective drug, ten years after Fleming had abandoned its development. They developed techniques for growing, purifying and manufacturing the drug, tested it for toxicity and efficacy on animals, and carried out the first clinical trials. In 1941, they used it to treat a police constable from Oxford. He started to recover, but subsequently died because Florey was unable, at that time, to make enough penicillin. Later trials in Britain, the United States and North Africa were highly successful. A graduate of the University of Adelaide, Florey studied at the University of Oxford as a Rhodes Scholar and in the United States on a fellowship from the Rockefeller Foundation. In 1935, he became the director of the Sir William Dunn School of Pathology at Oxford. He assembled a multidisciplinary staff that could tackle major research projects. In addition to his work on penicillin, he researched many other subjects, most notably lysozyme, contraception and cephalosporins.

Before the advent of nuclear reactors, protactinium was separated for scientific experiments from uranium ores. Since reactors have become more common, it is mostly produced as an intermediate product of neutron capture on thorium, used for the production of the fissile 233U:
== Contraindications == Zopiclone causes impaired driving skills similar to those of benzodiazepines. Long-term users of hypnotic drugs for sleep disorders develop only partial tolerance to adverse effects on driving, with users of hypnotic drugs even after one year of use still showing an increased motor vehicle accident rate. Patients who drive motor vehicles should not take zopiclone as there is a significantly increased risk of accidents in zopiclone users. Zopiclone induces impairment of psychomotor function. Driving or operating machinery should be avoided after taking zopiclone as effects can carry over to the next day, including impaired hand-eye coordination. A double-blind study on the effect on performance of several hypnotic medications, relevant to military personnel who may have to be awakened to carry out duties, found that drugs listed in increasing order of performance impact duration were melatonin (with no impact), zaleplon, temazepam, and zopiclone. The effects on serial reaction time (SRT), logical reasoning (LRT), serial subtraction (SST), and multitask (MT) were measured. For zaleplon (10 mg), zopiclone (7.5 mg) and temazepam (15 mg) respectively the times to recover normal performance for SRT were 3.25, 6.25, and 5.25 hours; for LRT 3.25, >6.25, and 4.25 hours; for SST 2.25, >6.25, and 4.25 hours; and for MT 2.25, 4.25, and 3.25 hours. The study did not consider the effectiveness of the drugs on sleep.

On 29 September P.W. Botha added a third objective to Operation Moduler: the destruction of all FAPLA units east of Cuito Cuanavale. The reasons for this shift in objectives once FAPLA had abandoned its offensive were not apparent to everybody in the South African government. Pik Botha and his senior colleagues in the foreign ministry cautioned against a major offensive north of the Lomba, citing potential diplomatic repercussions. But confidence in the SADF had been buoyed by its effective defence of the Lomba, and members of the South African general staff successfully agitated for a renewed offensive towards Cuito Cuanavale. It is unclear whether they interpreted their new objective as veiled permission to seize Cuito Cuanavale itself, although the option was discussed. Per Botha's new directive, the SADF commenced Operation Hooper with the goal of encircling the retreating Angolan brigades and preparing for operations further east of the Cuito River. The decision to commence Hooper towards the end of the 1987 calendar year created problems for the SADF, since a number of white conscripts involved in the Lomba River engagements were nearing the end of their national service. This led to a delay of several weeks while the existing troops were gradually withdrawn from Angola and replaced with a new intake. The SADF had dispatched a second mechanised battalion, 4 South African Infantry, to Angola, as well as a squadron of Olifant Mk1A tanks and a battery of G5 and G6 howitzers.

It is a synthetic cyclic heptapeptide and an analogue of alpha-melanocyte-stimulating hormone. The molecule is produced by chemical synthesis rather than extracted from a biological source.
It is shorter than the native hormone and carries a cyclic constraint that improves stability. These changes raise receptor potency and slow breakdown relative to the naturally occurring peptide.
Published accounts date its development to the 1980s, when researchers were generating analogues of melanocyte-stimulating hormone. That work aimed to produce more stable compounds for studying pigmentation biology.
It is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, containing seven amino acids with a lactam ring and a D-configured phenylalanine residue. It is supplied as a lyophilised powder for laboratory research.
