The first natural extractions were done with ethanol and afforded a 7:1 yield of crude extract to phytochemical; however, this technique produced a mixture of hyperforin and adhyperforin. The extraction technique has since been modernized using lipophilic liquid CO2 extraction to afford a 3:1 crude to phytochemical extraction which is then further purified away from adhyperforin. This CO2 extraction is rather tricky still because typical 'supercritical' conditions extract less material whereas anything over 40 °C (100 °F) will degrade hyperforin. It featured a weapon that created barriers and ramps from ice, and a blob-like enemy that could divide itself, consume other enemies and pass through grates. A leak of Valve development files in 2026 included a model for the Weaponizer, a weapon used to turn objects into ammunition. In 2009, reports surfaced that Valve was working with sign language and on a deaf character. Newell said that Gordon's companion, Alyx Vance, had programmed her pet robot, Dog, to use sign language, inspired by a deaf person she had a crush on. In 2010, Newell spoke of "broadening the emotional palette" of the Half-Life series, and said the next game may return to "genuinely scaring the player". According to Newell, whereas the original Half-Life (1998) saw the mysterious G-Man transform the protagonist, Gordon Freeman, into his tool, and Half-Life 2 saw Freeman being used by G-Man, the episodes would see G-Man lose control.

=== Offline combustion and reduction === Conversion to simple molecules (i.e. H2 for hydrogen) is required prior to IRMS for stable isotopes. This is for several reasons with regard to hydrogen: The classical offline preparation for the conversion is combustion over CuO at >800°C in sealed quartz tubes, followed by the isolation of resulting water and the reduction to H2 over hot metal at 400 ~1000°C on a vacuum line. The produced gas is then directly injected into the dual-inlet mass spectrometer for measurement. The metals used for reduction to H2 includes U, Zn, Cr, Mg and Mn, etc. U and Zn had been widely used since the 1950s until Cr was successfully employed in the late 1990s. The offline combustion/reduction has the highest accuracy and precision for HIC measurement without limits for sample types. The analytical uncertainty is typically 1~2‰ in δD. Thus it is still used today when highest levels of precision are required. However, the offline preparation procedure is very time-consuming and complicated. It also requires a large sample (several 100 mg). Thus, online preparation based on combustion/reduction coupled with the subsequent continuous flow-IRMS (CF-IRMS) system has been more often used nowadays. Chromium reduction or high temperature conversion are the dominant online preparation methods for detection of HIC by IRMS.
The wide-spread interest in the very remarkable specimen of the giant squid, now lying on the beach a few miles below the city, is mainly due to its enormous size. == Prognosis == The prognosis for alpha thalassemia depends on the degree to which alpha globin production is affected. Those with mild alpha thalassemia, involving deletion of one or two alpha-globin genes, do not generally require treatment and have a normal life expectancy. Hemoglobin H disease, with three of the four genes either deleted or inactive, gives a mild to moderate form of anemia but may lead normal lives. The prognosis when all four genes are affected, leading to Hb Bart's hydrops fetalis, is very poor, with most affected fetuses dying in utero or shortly after birth due to severe fetal hypoxia.
