The short version of afamelanotide fits in a sentence. The long version — which is the one that helps — is below.

Reversed-phase high-performance liquid chromatography is the routine method for purity assessment. Peptides absorb near 214 nm because of the peptide bond, and a gradient of acetonitrile in water separates the intact peptide from deletion sequences, oxidised products, and earlier-eluting fragments at neutral pH. Electrospray ionisation mass spectrometry provides an orthogonal check: the measured mass must agree with the theoretical value. Amino acid analysis and peptide mapping confirm structure but are used less often. Reference standards remain scarce because the peptide is not described in any pharmacopoeia.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.

Reported observations after unregulated use include shifts in skin pigmentation and, in some accounts, unintended changes to moles and other lesions. Whether these outcomes are causally linked to the compound, and how often they occur, remain open questions because controlled data are scarce. The absence of standardised dosing and verified product purity complicates interpretation. Researchers have called for better surveillance and analytical characterisation of samples obtained outside regulated channels. Conclusions drawn from anecdotal evidence should be treated as provisional.
Melanotan-2 has not received marketing authorisation from major regulatory agencies for any therapeutic indication. Several jurisdictions classify it as a prescription-only medicine or a controlled substance when supplied for human use. Because approved products do not exist, material sold online usually sits outside pharmaceutical supply chains and formal quality oversight. Regulators have issued public notices describing the compound as unapproved. Enforcement varies, and the legal position differs between countries, which complicates any single general statement about its status.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
As a result of the Partition of India in 1947, the formations, units, assets, and indigenous personnel of the Indian Army were divided between the Dominion of India and the Dominion of Pakistan. As Brian Lapping wrote, "By comparison with the two great provinces [Bengal & Punjab], partition of the army and the civil service was easy, though by any other standard, it was difficult, wasteful, and destructive. ... The men were transferred in their units. Regiments of Sikh and Hindu soldiers from the north-west frontier had to make their way through Muslim territory to get out of what was to be Pakistan." Also in 1947 a final agreement was signed regarding the Gurkha regiments in the British Indian Army. Four Gurkha regiments, recruited from both eastern and western Nepal, would join the British Army. The remaining six Gurkha regiments of the British Indian Army joined the Dominion of India. During the transition period after partition, those Gurkha regiments that were in Pakistan, did their service, but were eventually moved back to India. The partition reduced the ethnic imbalance of the British Indian Army, which became the present-day Indian Army. But, the partition resulted in more ethnic imbalance in the Pakistani military, mainly because the new nation state of Pakistan was formed by joining West Punjab, NWFP, East Bengal, Baluchistan, and Sind. The new Pakistan Army was mainly made up of soldiers from two of these provinces. The Bangladesh Army, which was created from the Pakistan Army on the independence of Bangladesh, retain many British Indian Army traditions.
=== Physical methods === Heat treatment − Treating food with heat, such as blanching or roasting, de-naturates enzymes and destroys the reactants responsible for browning. Blanching is used, for example, in winemaking, tea processing, storing nuts and bacon, and preparing vegetables for freezing preservation. Meat is often partially browned under high heat before being incorporated into a larger preparation to be cooked at a lower temperature which produces less browning. Cold treatment − Refrigeration and freezing are the most common ways of storing food, preventing decay. The activity of browning enzymes, i.e., rate of reaction, drops in low temperatures. Thus, refrigeration helps to keep the initial look, color, and flavour of fresh vegetables and fruits. Refrigeration is also used during distribution and retailing of fruits and vegetables. Oxygen elimination − Presence of oxygen is crucial for enzymatic browning, therefore eliminating oxygen from the environment helps to slow down the browning reaction. Withdrawing air or replacing it with other gases (e.g., N2 or CO2) during preservation, such as in vacuum-packaging or modified atmosphere packaging, wine or juice bottling, using impermeable films or edible coatings, dipping into salt or sugar solutions, keeps the food away from direct contact with oxygen. Impermeable films made of plastic or other materials prevent food being exposed to oxygen in the air and avoid moisture loss.
Pseudomonas syringae pv. tabaci, the causal agent of the wildfire of tobacco P. syringae pv. coronafaciens P. syringae pv. garcae P. syringae BR2, which causes a disease of bean (Phaseolus vulgaris) similar to tobacco wildfire. This organism is closely related to P. syringae pv. tabaci but cannot be classified in the pathovar tabaci because it is not pathogenic on tobacco. Tabtoxin is a dipeptide precursor to the biologically active form of TBL, differing by having an extra threonine attached by a peptide bond to the C terminus. Tabtoxin is required by BR2(R) for both chlorosis and lesion formation on bean. All mutations that affected tabtoxin production, whether spontaneous deletion or transposon-induced, also affected lesion formation, and in all cases, restoration of tabtoxin production also restored pathogenic symptoms. Other factors may be required for BR2 to be pathogenic on bean, but apparently these are in addition to tabtoxin production. This toxin is a irreversible inhibitor of glutamine synthetase.
Hays higher than 10% NSC may be used, but it is recommended to soak it for at least one hour before feeding in cold water, which can help reduce NSC levels, although this method is not always a reliable way to adequately drop them to acceptable levels. Obese horses are usually fed hay at a level of 1.5% ideal body weight, which may be dropped to 1% of body weight if no weight loss is realized after 30 days. However, feeding less than 1% of body weight in forage is not recommended, since secondary problems such as hyperlipemia and stereotypies can occur, and insulin resistance may actually be worsened. Removal of concentrates from the diet may be sufficient to produce weight loss in obese horses. Hays are often low in vitamin A, vitamin E, copper, zinc, and selenium. A vitamin and mineral supplement is added to the diet to ensure adequate nutrition. Ration balancers, which are low in calories but offer protein, vitamins, and minerals, are often recommended.
=== RIPA (RadioImmunoPrecipitation Assay) lysis buffer === RIPA buffer is a commonly used lysis buffer for immunoprecipitation and general protein extraction from cells and tissues. The buffer can be stored without vanadate at 4 °C for up to 1 year. RIPA buffer releases proteins from cells as well as disrupts most weak interactions between proteins. Recipe:

=== Energy === Formic acid can be used directly in formic acid fuel cells or indirectly in hydrogen fuel cells. Electrolytic conversion of electrical energy to chemical fuel has been proposed as a large-scale source of formate by various groups. The formate could be used as feed to modified E. coli bacteria for producing biomass. Natural methylotroph microbes can feed on formic acid or formate Formic acid has been considered as a means of hydrogen storage. The co-product of this decomposition, carbon dioxide, can be rehydrogenated back to formic acid in a second step. Formic acid contains 53 g/L hydrogen at room temperature and atmospheric pressure, which is three and a half times as much as compressed hydrogen gas can attain at 350 bar pressure (14.7 g/L). Pure formic acid is a liquid with a flash point of 69 °C, much higher than that of gasoline (−40 °C) or ethanol (13 °C). It is possible to use formic acid as an intermediary to produce isobutanol from CO2 using microbes.
=== Trump administration === On the same day of Powell's announcement, Trump denied having any knowledge of the investigation; White House Press Secretary Karoline Leavitt asserted that Trump did not order the Justice Department to investigate Powell. After suggesting at press conferences and social media posting in August and December 2025 that the administration would sue Powell due to the cost overruns, and after Powell announced that he was being targeted with an investigation, Trump said that Powell "either is incompetent or ... crooked" because of the cost overruns, and said that Powell will "be gone soon." However, Trump also said the investigation would not affect the timing of appointing a new Federal Reserve Chair as a successor to Powell. In an interview with Reuters on January 14, Trump said that he has no plans to fire Powell and that it was "too early" to say whether he would have legal grounds to remove Powell. Trump also dismissed concerns that diminished Federal Reserve independence would lead to higher inflation, asserting that "A president should have something to say" about monetary policy, while criticizing Republican Senators who may oppose his future Federal Reserve nominations due to the investigation, calling them disloyal. Secretary of the Treasury Scott Bessent has reportedly expressed concern that the investigation will cause volatility in the markets. While unnamed sources within the White House have suggested Federal Housing Finance Agency Director Bill Pulte may be behind the investigation of Powell, Pulte has publicly denied the allegations.
The attachment of fluorescent group on one head of the molecule confers contrast properties. A new strategy to enhance the cellular up-take capacity of CPP is based on association of polycationic and polyanionic domains that are separated by a linker. Cellular association of polycationic residues (polyarginine) with negatively charged membrane cells is effectively blocked by the presence of polyanionic residue (poly-glutamic acid) and the linker, which confer the proper distance between these two charged residues in order to maximize their interaction. These peptides adopt hairpin structure, confirmed by overhauser effect correlation for proton-proton proximities of the two charged moieties. At this stage only the linker is exposed to protease hydrolysis in vivo applications. The linker hydrolysis occur and the two charged fragments experience more conformational freedom. In the absence of linker, the cationic peptide can interact more efficient with the target cell and cellular uptake occurs before proteolysis. This strategy found applications in labeling tumor cells in vivo. Tumor cells were marked in minutes. Linker degradation can be predicted by the amount of D-aminoacids (the unnatural isomer) incorporated in the peptide chain, this restricts in vivo proteolysis to the central linker.
On a milligram consumed per kilogram of body weight basis, simian non-synthesizer species consume the vitamin in amounts 10 to 20 times higher than what is recommended by governments for humans. This discrepancy constituted some of the basis of the controversy on human recommended dietary allowances being set too low. However, simian consumption does not indicate simian requirements. Merck's veterinary manual states that daily intake of vitamin C at 3–6 mg/kg prevents scurvy in non-human primates. By way of comparison, across several countries, the recommended dietary intake for adult humans is in the range of 1–2 mg/kg.
This self-regulation effect is achieved as the CO2 laser essentially stops ablating as soon as it comes in contact with the intraocular percolated liquid, which occurs as soon as the laser reaches the optimal residual intact layer thickness.

Low temperature slows hydrolysis and oxidation, the two main routes by which the peptide backbone and side chains are modified. A lyophilised powder stored at −20 °C is more stable than one kept at room temperature, and once the material is dissolved the degradation rate rises, making refrigeration more important.
Liquid chromatography coupled to mass spectrometry is the most common approach because it combines a retention time with a mass measurement. A chromatographic peak alone cannot establish which peptide is present, so mass determination or amino acid analysis is used as an orthogonal confirmation alongside the separation.
This depends on buffer, pH, and concentration, and published stability data for this peptide are limited. In practice laboratories work through a refrigerated solution within a few weeks and discard samples that show cloudiness or visible particles. Freezing and thawing repeatedly is generally discouraged.
It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
